RESUMO

It was shown using the model of L2-dependent proliferation of T-cells that IL2 forms complexes with gamma-globulin inhibiting IL2 activity. In this interaction IL2 acts as lectin binding with two-antenna terminals of 5 or 6 mannose residues in one and the same region of immunoglobulin. Similar effect is observed in vivo in case of induction of DTH to listerial antigens. This phenomenon accounts for suppression of the immune response after passive injection of autoantibodies and for the inverse relationship between humoral and cellular immunity. Reduction of IL2 activity after its interaction with IgG or immune complexes may be regarded as a universal feedback immunoregulatory mechanism. External and internal factors showing affinity to mannose, e.g. sodium polyprenyl phosphate or lactobacillus, can interfere with this process and exert immunomodulating effect.

RESUMO

AIM: Demonstration of the ability of native products of lactobacteria to bind mannose containing N-glycans as a display of one of the mechanisms of probiotic behavior of these symbiont microorganism in gastrointestinal tract (GIT). MATERIALS AND METHODS: Lactobacillus plantarum 30 grown on selective medium (MRS-agar) and their ultrasound lysates were used in the study. Standard technique of delayed type hypersensitivity (DTH) reaction was used with inactivated Listeria monocytogenes culture as a priming agent. RESULTS: DTH reaction in vivo has demonstrated that oligomannoside N-glycan of egg albumin is a general acceptor for adhesins that are present in the native preparations of certain L. plantarum strains, as well as for phosprenyl (PHP) immunomodulator--an inhibitor of cell IL-2 reception. CONCLUSION: The data obtained give evidence that mannose specific adhesins that are conserved in lactobacteria preparations have the same binding sites in egg albumin as PHP and, therefore, IL-2.

RESUMO

The role of helper T-lymphocytes in the modulation of humoral immune response to sheep red blood cells with choleragen has been studied in vivo, the populations of cooperating marrow cells, formed in mice under the action of hydrocortisone being used as a model. In adoptive transfer marrow cells, taken from mice on day 12 after thymectomy and from mice previously treated with antithymocyte serum, have proved incapable of humoral immune response. Choleragen, similarly to theophylline, normalizes the humoral immune response of marrow cells in thymectomized mice, but inhibits this response in intact ones, while thymosin fraction 2 restores it again, thus abolishing the action of choleragen and theophylline. The opposite effects rendered by choleragen and theophylline on humoral immune response, depending on the hormonal status of the animals and the possibility of influencing these effects by means of thymosin fraction 2 indicate that the population of helper T-lymphocytes are selectively sensitive to changes in the concentration of intracellular cAMP. Their capacity for cooperative interaction in the immune process is regulated by thymic hormones and forms the basis of the mechanism permitting the modulation of humoral immune response with choleragen.

In the conditions of experimental deficiency of serum thymic factor (FTS) theophylline (TP) recovers in the post--thymic precursors of helper T lymphocytes an ability for differentiation to the effector regulatory stage and inhibits this process in the normal conditions. In the same conditions fraction 2 of thymosin (F2T) recovers this process and in combination with TP can relieve its opposite effects. The diversity of action of F2T on the differentiation of the post--thymic precursors of helper T lymphocytes is accounted for by the possibilities of realisation of the F2T and STF hormonal signals to these cells which depend on the condition of their specific receptors controlled by intracellular cAMP levels.

When studying the levels of immune response to the sheep erythrocytes in the adoptive transfer system in a population of cooperating bone marrow and spleen cells of the intact mice formed under the effect of hydrocortisone, differences have been found in their capacity to produce antibody-forming cells. Unlike the population of spleen cells formed during ontogenesis, the population of cooperating bone marrow cells acquires the capacity to develop an intensive enough immune response only after twofold immunization of the recipient and remains insensitive to the priming treatment of the donors with the sheep erythrocytes. These behavioral peculiarities of the population of cooperating bone marrow cells are due to relative deficiency of mature T-B helpers; the primed spleen T-lymphocytes are not involved in the formation of the pool of the latter.
